An overture is a technically a prelude, a prelude is just a short piece of introductory music that comes before a longer piece. In pre-baroque times the terms could really be used interchangeably. Over the years the term overture came to mean simply "the music that plays before the curtain rises" In modern times we usually take prelude to mean:
"a short introduction to an instrumental piece"
and Overture to mean:
"an instrumental work, that comes before a dramatic vocal work (opera or musical), and usually contains vocal themes of the piece that is to follow (kind of like a medly of the Musical/Opera).
For example, in the overture to "Phantom of the Opera" you'll hear "Phantom of the Opera", "All I Ask of You", "Music of the Night", etc.
